Once they are designated as such, they are prohibited from practicing with the team. They can, however, rehabilitate and participate in team meetings. If a player begins training camp on the PUP list, they can be moved to the active roster at any time, even after one practice. He does not have so many skills to be useful to the team without intimate knowledge of playbook. Besides on an injury settlement, the player can not come back until at least week 10. He can sign with another team though anytime, I believe, just not the team that waives him. That is also what happened with Christie a couple years back.
